High dose dietary vitamin D allocates surplus calories to muscle and growth instead of fat via modulation of myostatin and leptin signaling

In mice, and not peer reviewed yet, but very interesting.

"We used three levels of vitamin D in the context of defined CHOW VDRKR diets: 0 IU/kg (low – ENVIGO #140078), 2000 IU/kd (normal - ENVIGO #140079) and 10,000 IU/kg (high - ENVIGO #140080)."

Equivalent doses for an 80 Kg human: 16,000 to 80,000 IU.

I reckon i need to up my dose then. For years i took 5000 iu / day, but recently (month or so before hip hip replacement) i increased that to 10,000 iu. My last blood test on 5000 iu (and i was sunbathing too, but keep in mind i live in the UK) was 92 nmol/l (37ng/ml). I've read a lot about how higher levels can help with autoimmune conditions - i've had psoriasis since childhood, fairly mild now, but still get flare ups. I've also had occassional rheumatoid arthritis flare ups in my thumb joints over the years, and have it currently. Since increasing my vit d3 dosage to 10,000 iu, it hasn't done much for my thumb (just right hand at the moment). But, the small patches of psoriasis i've had on my left forearm, and one on the back of my scalp, for probably 2 years plus now, that steroid creams have not touched - have completely gone. Could be coincidence, but the only change i've made to my diet and supplementation has been with vit d3 dosage. Recently i've been reading a blog by a doctor that treats his patients with autoimmune problems with 30,000 iu a day for multiple years, with apparently great results and no side effects. Not sure i'm confident enough to go that high yet, but recent studies are showing that the fear of side effects from very high dosages are unfounded. Here's one example -

As a follow on from above - it's apparently important when taking vitamin d3, especially higher dosages, to also take additional vitamin A, vitamin K2, and magnesium. The vit A increases vit d's effectiveness, and vit K2 directs calcium the right way.

at 813IU/kg they reached above 30ng/dl. pretty vague. i reach 30ng/dl at 1k total.

That's a fairly low dose to reach that level (and i'm assuming you meant ng/ml, not dl). I guess that's why blood work's always important - individual variation, plus, there's always the issue of supplement stated dosage accuracy, unless it's pharma grade.

yes ng/ml ofc. at 10k IU I reach 80ng/ml, at 5kiu 60ng/ml consistently via blood test. so again, 30ng/ml does seem strange as in 80k/iu daily would put me at 200-300ng/ml. i tried once to run 50kiu/d and was hovering around 150ng/ml.

Wow, 50k iu a day! What were your reasons for such a high dose, were you trying to resolve an autoimmune type issue? I have been tempted recently to increase my own to 30k iu, just to see what, if any, effect it has on my rheumatic thumb joint. But i'd want to do blood work to see where it puts me, and at the moment, getting that done would be difficult (having just had my hip done, plus it's expensive).

no specific reasons, self experimentation something I am good at lol (and burned myself couple times). i felt no difference. been taking 10k for about a decade, and recently lowered it to 5k. also no difference. my focus has been on full body sun exposure lately, with 1h at noon in the pool daily at 8-10UV index (Arizona). i don't get burned but beyond 1h i start to get very exhausted despite pool cool-down, as it is still in the 100's currently. that level of sun ramps your D level pretty high and my understanding is that sun produced D is superior to supplements

'Self experimentation', yes, i've done a fair amount of that myself over the years too, lol. And i agree with you regarding the sun being a superior way to get your vit d (and according to Jack Kruse who i follow on IG, many other benefits too - i just wish i understood what the hell he's on about most the time). Unfortunately for me, living in the heart of the UK, i don't often get the kind of weather that helps build decent vitamin d levels.
